Transmission questions

So my cooler lines have developed a leak on my 05 2500HD. So I got the new lines and I was wondering will It drain the fluid out of the tranny while I replace the line? What is the best fluid to run or what is the stock fluid to run in the 4l80e? How much?

It will not drain all fluid. I use synthetic dexron 6 from Mobil 1
