The show Mythbusters on the Discovery channel addressed this for autos, and again for trucks in general.

To save gas in a pickup, swapping out your tailgate with a net yielded a %5 increase in gas mileage. Removing the tailgate by itself, lowering the tailgate, adding a tonneau cover, all had little effect or even decreased gas mileage.

As for additions the engine itself, nothing worked. The only thing you can add to your engine to affect your gas mileage is ethanol, and it will decrease your gas mileage as well as your horsepower.

I'm not sure of the 5% is valid.

I've read reports that with the tailgate up and no bed cover, an air bubble develops in the bed and the air coming over the cab is deflected by the bubble creating a smooth air flow.

Tests have shown that tailgate down DOES reduce MPG.

Royal purple is no gimmick... Many people, including myself, use it in everything. Engine, differentials, tranny, radiator... I know a lot of people that swear by it. I'm not saying it will increase your fuel mileage, but I wouldn't be surprised.

Both my trucks gained 2-3 MPG (depending on driving situations) when I swapped out the fluids for Royal Purple Synthetics. See my Signature below.

I saw this episode a few days ago, they hooked a flow meter to the fuel delivery system to show fuel consumption using different scenarios. Tailgate up, Tailgate off, Hard Tonneau cover installed all showed similar results on the meter. Tailgate down proved to be the worst configuration while no Tailgate with a net showed to be the best.
