Is there any gain by using 3/4"MDF over 1/2"MDF?

I have noticed most people use 3/4.

Yes, it is much more rigid.  If you are reinforcing with fiberglass, you can use the 1/2 inch though for anything less than 2 15's.

It has to do with resonance.  A lot less with 3/4".  You can use 1/2" and try 75% fill or try some dampner pads designed for sub enclosures.

To clarify my statement above.  Your box can never be TOO Stiff.  I have always used 3/4 MDF or 3/4 Marine Plywood with fiberglass reinforcement for all sub enclosures.
