Yes, it is much more rigid.  If you are reinforcing with fiberglass, you can use the 1/2 inch though for anything less than 2 15's.

To clarify my statement above.  Your box can never be TOO Stiff.  I have always used 3/4 MDF or 3/4 Marine Plywood with fiberglass reinforcement for all sub enclosures.

 

Systems built with 3/4" MDF:

2 8" -Mitsubishi Talon

4 8" -S10

8 8" -Accord

2 10" -Cherokee

2 10" -Mustang

4 10" - CRX

4 15" - Bronco

8 18" - Mazda PU bed
