Correct Method Of Getting Fuel Pressure Reading?

Just a question about getting a fuel pressure reading on the rail.

 

Is the correct method to get a reading from the test gage with the Fuel Pressure Regulator line connected or disconnected?

 

The Yukon beast has 50psi at the rail with the key in the ON position, but not running, 45 psi running, and 55 with the vacuum line off the FPR.

 

Any ideas?

My '01 silverado has about 52psi key on engine off, about 48-50 running, and about 60 running with fuel pressure regulator vacuum line disconnected, runs fine now that I just had the clogged cats replaced.
