On my 2014 Sierra 1500 with a 5.3 L83 PO306 Spark is good but compression is 90 PSI... Would it be okay to recheck the compression with the lifters on #6 removed? will it hurt anything?

Well you can't check compression if the lifters are removed because the cylinder heads have to come off for that. Did you mean rocker arms?

 

I'd just pull the valve cover to see what you find on that cylinder. If the lifter is collapsed on cylinder 6, you'd know pretty quick.
