Can a cylinder 2 misfire be a lifter if not d.o.d.?

We have a 5.3 misfire on a non displacemtn on demand cylinder #2 that we cannot fix. Is it possible that it is a lifter if it is not d.o.d.? We have tried everything and don't want to change lifters and find out it does not solve anything.

Cannot fix as in a misfire on #2? Is the lifter clacking? Did you do a compression check? Did you have a lean code at any point? How many miles on the engine? Do you have any other codes? If you don't have lifter noise, check compression. A couple of possibilities can be a leaking head gasket or burnt valve. Fuel injector good?

Well if you have done everything then you are done. Probably best to just drop in a new long block. Pace performance and several others regularly run specials on rebuilt 5.3s around 3k BAM done. I guess it is according to what truck you have (might be a 500 dollar beater LOL) but for 3 grand it might be worth a look.

135k miles, spark, coil, smoke test good, literally every test done. When you drive it it drives good and then suddenly starts shaking and driving rough with a cylinder 2 misfire. Compression is down when this happens otherwise good. Appears to be some minor scratches in the offending cylinder not found in cylinder 4.
