I have a 2016 gmc sierra 2500 with 6.0 vortec n 4.10 rear. I find it easier to tow my travel trailer when 6th gear is locked out. Is this OK for me to do?

Yes it should be fine. I assume you have it in manual set to 5th gear.

I just use the tow/haul setting and my 6.2l, 6 speed, 3.73, and it tows awesome because it keeps it at the right rpm where it has the most torque/power for the gear.

Did that all the time in my '15 Sierra 1500. While towing my 8500lbs travel trailer especially. There were times in the more hilly rural hwys with 55mph speed limits that i'd leave it in 4th even. Kept the power up and wasn't hunting gears all the time. Made it much more pleasant to drive.

Of course I have since recently traded up to a diesel to solve that problem. :)

Yep, as stated, it's just fine to tow in M5. Using tow/haul should keep it from hunting gears as much, but if it still does, then locking out the higher gears is better. You'll probably even find that you tranny temps will be lower in M4 or M5 than having it shift all the time.
