I have a 2016 Chevy 1500.  I just rolled over 123XXX miles.

Just recently these past couple of days, when on hire, noticing some hesitation.

Around 35-60mph there is hesitation, lack of acceleration, jumping, etc. - but only if i am maintaining speed.

If i go into a heavy acceleration or slow down, everything is fine.  This only happens in V8 mode, but if the AFM knocks it down to V4, no issues at any cruising speed.

 

Also the other day, I noticed from a stop - between 1st - 2nd- 3rd gear shifts, there seems to be a slight lag with a whinny hum, but only if I am driving slowly and ramping up slowly.

 

Not sure where to start with diagnostics, or if I should just take it in.

Well, are you going to fixing it or just taking it in is the first question. 

Second question, did you scan it for stored codes?

Third question, was proper maintenance done?

Kind of depends on what the issue could be. 
Proper maintenance has been done.  Did not store for codes. 
As mentioned - was not sure where to necessarily start.  I’d be fixing it if it’s found to be something in my wheelhouse.

Some of that sounds like a Torque converter issue, try driving in manual mode 5th gear on the highway and see if it does the same thing.

I had what sounds like a similar issue with my 2016.  Codes indicated a bad injector which I replaced with some money from GM's "we feel bad about this" fund. Problem gone
