If you plan to keep the truck more than 100k miles, change the transmission fluid at 50k miles. If you plan to sell or trade it before 100k, then save your money and skip the fluid change. It will run about $350-$400 based on what I paid on my GMT900 truck.

Unless you are towing a lot the factory service interval on the 6L80E is 100k miles. I have no intentions of checking mine before then and didn't on my prev 6L80E vehicles.

I wouldn't follow that guideline at all. The manufacturer most likely recommends 100k to keep you from doing any work to the tranny during the warranty period. Conventional wisdom says to change transmission fluid around 50k (30k if you do a lot of towing). If you reach 100k without ever changing it, it's better to leave it alone at that point than to change it because you will flush out the metal shavings and bits that have worked their way into the seals in the transmission and actually create leaks and transmission problems.

That has always been my mindset as well. I've known several people who have gone years without transmission maintenance only to get suckered in by their dealer or local mechanic for a trans fluid flush, and next week what do you know? Transmission is slipping now and it needs to get re-built.

I will never do, nor have ever "flushed" a transmission ever. I worked in a transmission rebuild factory for a year, one of the things I learned that flushing is the worst thing you can do.

 

If you want to change early go ahead, the 6L80E has a specific procedure in order to change the fluid, just make sure you follow the fill procedure. On my G8 GXP it was a pain since they didn't have a fill tube and you had to do it from under the car on level ground and while up to temp.

I'm a believer in the thinking of either: Do it early and often, or don't do it at all. My 99 Sierra has over 120k miles on it and the only thing I've ever done to the transmission is check fluid level occasionally. My dad has an 89 Chevy with over 230k and the transmission has never been serviced. How a 700R4 has done that is a mystery to many. I've heard several people start having issues if they changed fluid and filter after 100s of 1000s of miles, just as one of the above posters mentioned.

It is important to note there's a difference between a FLUSH and a fluid change, sucking out the fluid then shoving clean fluid in then sucking it out again and topping it off to finish off a "service" is where you wind up causing metal shavings and debris to dislodge and cause issues,

 

Draining the transmission and refilling using a new filter I don't see that as something to shy away from w/ 50,000 miles or 250,000 miles
